This MAb reacts with MUC1, a large transmembrane glycoprotein expressed on the ductal surface of normal glandular epithelia. It is used as tracer agent in CA15.3 assays. The extracellular domain of MUC1 largely consists of a highly conserved, O-glycosylated 20 amino acids tandem repeat which can occur 30-100 times per molecule depending on the length of the allele involved. In the vast majority of human carcinomas this protein is up-regulated and poorly glycosylated and appears on the cell surface in a non-polarized fashion. The dominant epitope of this MAb involves both amino acids as well as sugar moieties. Cadherin 17, also known as LI cadherin (for liver-intestine cadherin), is a member of the cadherin family of calcium-dependent adhesion molecules. Cadherins mediate cell-cell interactions via hemophilic binding, and are critical to the maintenance of tissue structure and morphogenesis. Cadherins each contain a large extracellular domain at the amino terminus, which is characterized by a series of five homologous repeats, the most distal of which is thought to be responsible for binding specificity. The relatively short carboxy terminal, intracellular domain interacts with a variety of cytoplasmic proteins, including beta-catenin, to regulate cadherin function. Cadherin 17 expression is restricted to liver and intestine tissues and is specifically localized to the basolateral domain of hepatocytes and enterocytes. The insulin receptor (INSR) is a heterodimeric protein complex that has an intracellular subunit, which is disulfide-linked to a transmembrane segment. The insulin ligand binds to the INSR and initiates molecular signaling pathways that promote glucose uptake in cells and glycogen synthesis. Insulin binding to INSR induces phosphorylation of intra-cellular tyrosine kinase domains and recruitment of multiple SH2 and SH3 domain-containing intracellular proteins that serve as signaling intermediates for pleiotropic effects of insulin. Type 1 diabetes is an autoimmune condition of the endocrine pancreas that results in destruction of insulin secreting cells and a progressive loss in insulin-sensitive glucose uptake by cells. S100A and S100B proteins are two members of the S100 family of calcium binding proteins. S100A is composed of an alpha and a beta chain whereas S100B is composed of two beta chains. This antibody is specific against an epitope located on the alpha-chain (i.e. in S-100A and S-100B) but not on the beta-chain of S-100 (i.e. in S-100B). This antibody can be used to localize S-100A in various tissue sections. S-100 protein has been found in normal melanocytes, Langerhans cells, histiocytes, chondrocytes, lipocytes, skeletal and cardiac muscle, epithelial and myoepithelial cells of the breast, salivary and sweat glands. Neoplasms derived from these cells also express S-100 protein. ATRX is a member of the Snf2 family of helicase/ATPases, which contribute to the remodeling of the nucelosome structure in an ATP-dependent manner, and facilitate the initiation of transcription and replication. Structurally, ATRX contains a PHD zinc finger motif. ATRX is regulated throughout the cell cycle where it is differentially distributed within the nucleus. During interphase, ATRX predominately associates with the nuclear matrix, while during mitosis, ATRX localizes with condensed chromatin. At the onset of M phase, phosphorylation rapidly induces this redistribution of ATRX to the short arms of human acrocentric chromosomes, where it then specifically complexes with heterochromatin protein 1 thalassemia or ATRX syndrome. Focal adhesions are identified as areas within the plasma membrane of tissue culture cells that adhere tightly to the underlying substrate. In vivo, these regions are involved in the adhesion of cells to the extracellular matrix. Paxillin and vinculin are cytoskeletal, focal adhesion proteins that are components of a protein complex which links the Actin network to the plasma membrane. Vinculin binding sites have been identified on other cytoskeletal proteins, including talin and alpha-actinin each contain actin binding sites. Reportedly, vinculin is a potential plasma biomarker for Age-related Macular Degeneration (AMD). The early detection of AMD using novel plasma biomarkers with genetic modeling may enable timely treatment and vision preservation in the elderly.
